We were there last May.. Stayed at the Flamingo, mid strip..
Saw Mystere and thought it was great.. Bought front row ticks at a Kiosk in the Venetian. Paid much less for the tickets, but had to go that night.
Also saw Blue Man group, that was pretty cool as well!
If you get a chance, go the Fremont Street, great place to walk around and see some sights.. They had a Woodstock / 1969 light show on the "ceiling" this past year, probably not there now..
Do NOT go to VooDoo lounge at the Rio.. Very pricey, portions not that large, and our wonderful waiter stole $100.00 from the tab..My Two favorite teams are the Packers, and whoever plays the Vikings!

Been to Vegas about 15 times in last 8 years (it's only 3-4 hr drive for me).
Quick trip over to the Hoover Dam to get out of the city.
Check out the 'Pawn Shop' from the television show. If you haven't seen it - it's fricken hilarious.
Phantom was very well done.
The shark exhibit is worth the money (like $10-$15).
If you're smart, get her a day spa at one of the nicer hotels. Gives you a chance to see your 'adult' shows or just gamble solo. And it's worth it when she's all relaxed. :P
You can go to Mystere for under $100. Funny story - I went about 6 years ago and was randomly selected from the crowd to be a bit actor. They picked me up in a golf cart, ran me through a fake wall and immediately dressed me in a bib, diapers and bottle. I followed the FAT baby around and did some silly shit. But a good memory.
Then make a night of just seeing the strip - the light shows, ship fights, canals, shopping.
Personally I can't stand Vegas for more than 2 days. Too much drinking, craziness, etc.The measure of who we are is what we do with what we have.

We're headed there for 3 days and 2 nights; just a getaway. Heard Mystere was about 200 per person.
